  • Développements méthodologiques et innovations techniques:
    J’ai été recruté comme Ingénieur de Recherche au CNRS en 2004 venant d’un poste de Product Manager chez GVInstruments (ex-Micromass) fabricant de spectromètre de masse isotopique affecté à l’UMR5125 Paleo-Environnements Paleo-bioSphere (PEPS) dirigée par le Pr Lécuyer devenue l’UMR fusionnée 5276 en 2011 dirigée par le Pr Albarède puis par le Pr Mattioli depuis 2015. J’étais motivé pour intégrer cette structure car, historiquement, l’unité de recherches PEPS était réputée pour les analyses isotopiques de phosphates et carbonates utilisant des techniques de préparation « offline » avant analyse par spectrométrie de masse, qui ont été développées par le Pr Lécuyer. Les 3 premières années (contrat CDD) ont consisté à automatiser les systèmes de préparation en collaboration avec un autre IR (Dr F. Martineau). Ainsi, la première étape fut pour moi d’installer un système automatisant la préparation en ligne avec le spectromètre de masse des échantillons de carbonates et d’eaux pour les analyses 2H/1H, 18O/16O des eaux et 18O/16O, 13C/12C des carbonates. Ce système « MultiPrep » avait été mon projet de développement lors de mon arrivée chez Micromass en 1996. Il a permis d’augmenter d’un facteur 2 à 3 la précision des analyses, de diminuer la prise d’essai du milligramme au microgramme afin d’observer des variations à petite échelle et d’améliorer le rendement d’analyses permettant de développer plus de collaborations externes. Ce système est toujours en activité.
    La deuxième action significative fut l’installation et le développement des systèmes analytiques dits « en flux continu » par rapport aux systèmes traditionnels dits « en double introduction ». En effet, initiées dans les années 80 les techniques de « flux continu » ont connu une forte progression au cours des années 90, pour devenir un outil indispensable dans les années 2000 pour tout laboratoire utilisant les isotopes stables. Ces techniques permettent de produire plus rapidement et plus efficacement d’importants jeux de données isotopiques en conservant des performances de très haut niveau. En revanche les contraintes analytiques sont sensiblement différentes des techniques « en double introduction » ce qui a nécessité une réorganisation importante du laboratoire. La première étape fut l’installation d’un analyseur élémentaire négociée à 10% du prix catalogue avec la société GV-Instruments. Ce système a permis de réduire la prise d’essai nécessaire aux analyses 18O/16O de l’émail dentaire passant de 8 milligrammes à 500 microgrammes. ces analyses représentaient approximativement 80% des analyses isotopiques de l’UMR5125-PEPS. Cela a permis d’étudier les variations de paléo-saisonnalité de la température de l’air ou des eaux marines, ouvrant ainsi de nouvelles perspectives scientifiques dans le domaine des reconstitutions paléo climatiques.
    Le développement des analyses en flux continu a connu une nouvelle étape en 2010 avec l’établissement d’une collaboration avec la société Elementar-IsoPrime devenue le nouveau nom commercial de GV Instruments et spécialisée dans l’analyse élémentaire. L’objectif de cette collaboration était d’adapter le nouveau système « Pyrocube » présentant des innovations techniques significatives, aux contraintes des analyses isotopiques. Ce système d’un prix catalogue de 70Keuros a été mis à disposition gratuitement depuis 2010. Après une période de mise au point en collaboration étroite avec la société Elementar-IsoPrime et ses ingénieurs basés à Hanau (Allemagne), Manchester (UK), Lyon et Paris, ce système a permis une amélioration significative de la précision des analyses 18O/16O de l’émail dentaire ainsi qu’une réduction supplémentaire [cD1]de la taille de prise d’essai (c. 30microgrammes).
    A partir de 2011 ce même système a également été utilisé pour développer les analyses isotopiques simultanées de 13C/12C, 15N/14N et 34S/32S. Il s’agissait des premières analyses isotopiques 34S/32S effectuées à Lyon offrant là aussi de nouveaux champs d’investigation notamment dans le domaine de l’identification des environnements aquatiques (eaux douces ou marines) de tétrapodes très anciens à l’écologie très mal connue. Au-delà de ce thème principal, ce développement méthodologique a également permis le développement de collaborations interdisciplinaires fructueuses, l’une dans le domaine de l’archéologie en collaboration avec le musée des confluences de Lyon et celui de Las Palmas (Îles Canaries) ainsi qu’une dans le domaine médical en collaboration avec le centre hospitalier Lyon-Sud et le centre Européen de Nutrition Humaine de Lyon. Ce système, nommé ‘Pyrocube’, est devenu aujourd’hui la référence au niveau mondial dans le domaine des isotopes stables et est maintenant à la source de 80% des [cD2]publications de l’UMR ainsi que de diverses collaborations.
    Au cours de ces années, mon passé dans l’industrie a été utile afin de participer aux négociations avec les constructeurs pour les achats, tests et installations de deux spectromètres de masse avec périphériques l’un dans l’équipe « Vie Primitive » de l’UMR5125 (puis 5276) ainsi qu’un instrument installé dans l’UMR5023 « LEHNA ».
    Grâce aux compétences acquises pendant mes années chez un constructeur d’instrumentation parallèlement aux nouveaux instruments de mesure des isotopes stables, j’ai également été en charge de réhabiliter des appareils plus anciens récupérés dans d’autres laboratoires qui ne parvenaient plus à les faire fonctionner. Ceci a permis de répondre rapidement aux besoins analytiques grandissant du laboratoire. Quatre de ces appareils sont maintenant installés dans l’UMR5276 et sont à la disposition des étudiants et chercheurs invités. Deux sont stockés pour fournir des pièces détachées et deux sont en attente de réhabilitation. Ceci a été réalisé pour des coûts extrêmement réduits compte tenu de la qualité du matériel récupéré. Ceci a été possible grâce aux réseaux de contacts acquis lors de mes postes précédents Il est à noter que cette tâche de réhabilitation a été considérablement ralentie suite au départ en retraite de mon collègue IR Dr F. Martineau en 2013 sans remplacement. De plus les inondations dues aux travaux effectués par l’UCBL sur le bâtiment abritant ce matériel en juin 2015 (processus d’indemnisation bloqué) ont également touché certains de ces appareils anciens retardant encore leur remise à niveau. Le développement de ce partenariat public-privé m’a également donné l’occasion d’être invité en moyenne à une à deux fois par an à présenter les travaux de notre UMR lors des journées utilisateurs organisées par la société Elementar-IsoPrime.
    Un de ces systèmes en particulier a été réhabilité afin de pouvoir utiliser une technique originale de mesure des rapports D/H des eaux automatisée par réduction au chrome ayant permis de développer un projet fondamental en collaboration avec le Muséum d’Histoire Naturelle de Paris (Dr F. Robert) pour étudier le fractionnement isotopique de l’hydrogène lors des réactions de sublimation de l’eau pour diverses conditions de pression et température. Ceci nous a permis d’intégrer un groupe de travail qui contribue à la réalisation du projet LUNA21 de l’ESA (projet en cours) pour la conception d’un système d’analyse destiné à une mission lunaire.
    Depuis septembre 2015 je suis à l’origine de la définition d’un projet de plateforme analytique régionale pour les isotopes stables au sein de l’UCBL en collaboration avec l’UMR 5023 LEHNA et la fédération de recherche IFR41 (projet en cours).
    Au cours de ces années j’ai été amené à travailler avec 3 techniciennes successives différentes que j’ai formé entièrement à la spectrométrie de masse puis encadré, dont une travaille toujours avec moi depuis 2011.
     
    Implications dans les projets scientifiques et encadrements de doctorants et post-doctorants : 
    Depuis 2004 j’ai été amené à encadrer 13 doctorants (2 en cours) ainsi que 2 post-doctorants (1 en cours) ainsi qu’en moyenne 1 à 2 stages de M2 par an. De plus j’ai été sollicité pour participer à divers projets scientifiques comme les projets ANR DEEP, NAPCOD avec l’unité de recherche LEHNA ainsi que le projet ANR-08-BLAN-0227 ‘ACTES’ porté par Denis-Didier Rousseau de l’ENS Paris ou encore le projet ERC Advanced Grant PaleoNanoLife porté par F. Robert du MNHN Paris.
     
    Une étape importante dans mon projet de carrière : obtenir le grade de Docteur de l’Université Claude Bernard Lyon 1
     
    Face au besoin d’encadrement de notre équipe et de mon souhait de progresser dans le corps des IR, le Pr. Lécuyer m’a proposé un sujet de thèse portant sur le développement de l’analyse des isotopes du soufre dans la matière organique et surtout dans les apatites biogènes. Mes quatre années de travaux de thèse m’ont permis de publier 46 articles scientifiques que j’ai cosigné dont 4 en premier auteur et d’obtenir ainsi le grade de Docteur mention Sciences de l’Univers en Octobre 2014 devant un jury composé du Pr T. Fallick (Université de Glasgow-UK), Dr C. France-Lanord, DR CNRS (CRPG-Nancy), Pr C. Lécuyer (UCBL), Dr B. Malaizé, MC-HDR (Université de Bordeaux), Pr A. Mariotti (UPMC Paris), Dr B. Reynard, DR (CNRS ENS Lyon), Dr F. Tamburini, Chercheur (EPF Zürich-Suisse).
    Afin de continuer dans cette voie pour mon équipe et pour ma carrière j’envisage de préparer mon HDR dans le domaine des isotopes stables. Ce projet doit débuter en Octobre 2016 avec un développement analytique majeur en collaboration avec la société IsoPrime-Elementar qui doit installer en prêt à Lyon un tout nouveau système analytique « IsoFlow » ainsi qu’un nouveau type de spectromètre de masse « PrecisIon » représentant une valeur catalogue de 200K€, dont nous serons les premiers à bénéficier et qui devrait nous permettre d’aller encore plus loin dans les investigations scientifiques de nos thèmes de recherche.
     
    Expertise reconnue : sollicitations à la formation et invitations à animer la vie scientifique :
    Depuis mon retour dans le domaine académique je suis devenu un expert sollicité en moyenne 2 à 3 fois par an par les éditeurs de revues scientifiques internationales telles que Journal of Mass spectrometry, Rapid Communication in Mass Spectrometry, G-cube. Je participe régulièrement à des congrès internationaux afin de présenter mes travaux; par exemple les congrès de la Société Française des Isotopes Stables - SFIS 2006, 2010, 2014; les congrès du Joint European Stable Isotopes Users Meeting - JESIUM 2008, 2016 ; les congrès de l’Applied Isotope Geochemistry - AIG 2011, 2015 ou le congrès de l’American Geophysical Union – AGU en 2013. Suite à ma présentation de l’AIG 2011 à Tarragona (Espagne) j’ai été invité par l’éditeur de l’ouvrage intitulé « Gas Source Isotope Ratio Mass Spectrometry» à participer au chapitre sur les isotopes stables : Willi A Brand, Charles B Douthitt, Francois Fourel, Rodrigo Maia, Carla Rodrigues, Cristina Maguas, Thomas Prohaska (2014) New Developments in Mass Spectrometry: Sector Field Mass Spectrometry for Elemental and Isotopic Analyses" Royal Society of ChemistryThomas Graham House, Science Park, Milton Road, Cambridge, CB4 0WF, UK.
     
    Au-delà de mes participations à des congrès internationaux, en 2007, j’ai obtenu et organisé les « journées jeune chercheur » de la Société Française des Isotopes Stables (SFIS) que nous avons organisées à Lyon avec l’aide du Pr Lécuyer et de Dr Martineau et qui reçurent un franc succès auprès des participants. Plus récemment en 2015 mon expertise a été reconnue pour participer au jury de concours de recrutement d'un IGE externe ingénieur en techniques d'analyse chimique pour la plateforme GISMO/SEDS-UMR Biogéosciences université de Dijon.
    Dans le domaine de la formation au niveau régional au sein de la DR7, depuis 2015 j’ai co-organisé une formation CNRS intitulée « Analyse des isotopes 2H, 13C, 15N, 18O, 34S dans les matrices organiques ou inorganiques par spectrométrie de masse des rapports isotopiques » en partenariat avec l’Institut des Sciences Analytiques de Lyon (Dr P. Jame UMR 5280). Une première formation a été assurée en mars 2016. De plus en plus impliqué dans le domaine de la formation, en septembre 2016 j’ai été invité à participer au projet PHC Siam en allant donner une série de cours à l’université de Mahasarakham (Thaïlande) puis en dispensant à Lyon une formation en isotopes stables pour deux chercheurs Thaïlandais de cette même université en novembre 2016. Enfin, depuis septembre 2016 mon expertise dans le domaine des isotopes stables a été une nouvelle fois reconnue par mes collègues puisque j’ai été élu membre du conseil d’administration de la SFIS.
